TLR agonist therapy induces persistent immunity. 105 CT26 cells were implanted into the flank of syngeneic BALB/c mice. Large established tumors (≈800 mm3 in volume) were treated as described in Figure 5. A) Cells from the tumor draining LN were isolated one day after the third treatment, stimulated ex vivo with AH-1 peptide, and monitored for IFNγ secretion by ELIspot assay. Results represent the mean + SD of 4 independently studied mice/group. B) Mice cured of their CT26 tumors by treatment with CpG ODN plus 3M-052 (a cure being defined as being free of detectable tumor for ≥2 months after the cessation of therapy) were re-challenged with 106 CT26 cells. Their survival compared to naive mice challenged with the same tumor dose is shown (N = 6 mice/group). *, p < 0.05; **, p <0.01, ***, p <0.001.
